Microsome Preparation
We offer The prepared microsomal fractions taken from a liver of humanized CYP3A mouse. We also offer microsomal fractions from mouse-origin CYP3A knock out mouse and wild type mouse as a control.
Microsome preparations method:
Extraction of the liver after blood removal treatment,

Homogenate the liver in 50 mM Tris-HCl (pH 7.4) that contains 2 mM EDTA 150 mM KCl.

Centrifugal separation(9,000×g)
↓
Supernatant collection (S9 fraction)
↓
Ultracentrifugal separation (105,000 × g)
↓
Resuspension of the precipitate using 50 mM Tris-HCl (pH 7.4)
↓
Ultracentrifugal separation (105,000 × g)
↓
Resuspension of the precipitate using 250 mM sucrose solution
↓
Storage at -112 °F
